As with the first Slay the Spire, your journey begins with the swords weilding Ironclad. You'll have to work your way through the Spire to unlock the next character - The Silent.
Whether you are defeated or choose to give up, you will unlock your next character.
The same applies for every other character in Slay the Spire 2: The Regent, The Necrobinder and The Defect.
The order in which they are available are as follows: Ironclad, The Silent, The Regent, The Necrobinder and The Defect.
